ABSTRACT

BACKGROUND: In the elderly patients with intertrochanteric fractures, a large amount of hidden bleeding appears after surgery, so reducing hidden bleeding has become an important problem to be solved urgently. OBJECTIVE: To evaluate the effect of preoperative intravenous injection of tranexamic acid on the hidden blood loss after proximal femoral nail antirotation implantation. METHODS: Eighty-two cases of senile intertrochanteric fractures at Department of Traumatic Orthopedics of Guang’an People’s Hospital from July 2018 to July 2019 were selected, and all the patients underwent internal fixation with proximal femoral nail antirotation. The patients were allocated into two groups based on usage of tranexamic acid (n=41/group). In trial group, intravenous injection of tranexamic acid (10 mg/kg) was conducted during anesthesia, and control group was given the same volume of normal saline via vein. Blood routine examination was conducted at baseline and 1, 3 and 5 days postoperatively to count the level of hemoglobin and hematocrit. The total blood loss was calculated by Gross equation. The transfusion rate and postoperative incidence of thrombosis were recorded. RESULTS AND CONCLUSION: (1) There was no significant difference in the dominant blood loss between two groups (P > 0.05). (2) The perioperative actual total blood loss and postoperative hidden blood loss in the trial group were significantly lower than those in the control group (P < 0.05). (3) Three cases of blood transfusion (7%) in the trial group, and eight cases (24%) in the control group (P < 0.05). (4) No thrombosis occurred in both groups. (5) These findings suggest that preoperative intravenous infusion of tranexamic acid can effectively reduce the hidden blood loss after proximal femoral nail antirotation implantation, and does not increase the risk of postoperative thrombosis.

ABSTRACT

ObjectiveIn order to compare the alteration of reelin-immunoreactive Cajal-Retzius cells (CR cells) in molecular layer of dentate gyrus of APPswe transgenic mice with wild type, the histochemical and developmental characteristics of CR cells were studied, therefore, the roles of CR cells in Alzheimer's disease would be revealed further.Methods The Thioflavine S staining, reelin immunofluorescence with or without reelin/glutamate and reelin/GABA immuno-double staining were carried out in the study. In the meantime, Western blotting was used to study the expression of reelin in hippocampi of the both wild type and transgenic mice. Results Reelin positive CR cells could be double-labeled with either glutamate or GABA immunostaining. Caspase-3 immunofluorescence demonstrated that some CR cells went through apoptosis during their development. Compared with wild type, CR cells in APPswe transgenic mice had significantly decreased in the molecular layer of the dentate gyrus. The result was supported with Western blotting analysis of reelin expression in hippocampus. Conclusion Reelin could be co-expressed with either glutamate or GABA, suggesting CR cells would be glutamatergic exciting neurons and GABAergic interneurons. The loss of CR cells during development probably was caused by the neuroapoptosis. Significant decrease of CR cells in hippocampus of APPswe transgenic mice indicated reelin may play an important role in AD pathological alterations.

ABSTRACT

The prenatal ethanol exposure induced the alterations of dendritic spine and synapse in visual cortex and their long-term effect would be investigated in mice from P0 to P30. Pregnant mice were intubated ethanol daily from E5 through the pup's birth to establish mode of prenatal alcohol abuse. The dendritic spines of pyramidal cells in visual cortex of pups were labeled with DiI diolistic assay, and the synaptic ultrastructure was observed under transmission electron microscope. Prenatal alcohol exposure was associated with a significant decrease in the number of dendritic spines of pyramidal neurons in the visual cortex and an increase in their mean length; ultrastructural changes were also observed, with decreased numbers of synaptic vesicles, narrowing of the synaptic cleft and thickening of the postsynaptic density compared to controls. Prenatal alcohol exposure is associated with long-term changes in dendritic spines and synaptic ultrastructure. The changes were dose-dependent with long term effect even at postnatal 30.

ABSTRACT

Objective To analyze the clinical effect of brevisapine injection on high sensitivity C-reaction protein level in the treatment of acute myocardial infarction.Methods The patients were randomly divided into treatment group(n=80)and control group(n=80).Both groups were treated with routine midication,but treatment group were given intravenous drip of 100 mg brevisapine once a day.for 14 days.80 paients who came for diagnosis were selected for healthy control group.The selqlm high sensitivity C-reaction protein levels were measured at 2 d,7 d,and 14 d by ELLSA in treatment group,control group and healthy control group.Mortality,heart function status (Killip class),revaseularization and clinical adverse cardic events including postinfartion angina,new arrhythmia,reinfarction and bleeding complication at 4 weeks were observed.Results Treatment group had a significantly high level of hs-CRP compared with healthy subjects[(6.37±1.43)vs.(2.17±1.12)mmol/L,P<0.01]at the second day.The serum hs-CRP content of treatment group was obviously lower than that of control group(3.21±1.31)and(2.25±0.34)mmol/L vs.(5.87±1.16)、(3.97±1.21)mmol/L,(P<0.01)]on the 7th,14th days;The revascularization rate of treatment group was obviously higher than that of control group[62.50%(50/80)vs.51.25% (45/80),P <0.01];There was no difference between the two groups in mortality(3.75% vs.4.10%,P >0.05).No difference was found in the rate of bleeding between the two groups(6.25% vs.7.50%,P >0.05);Patients with Killip class<Ⅱ of treatment group was obviously higher than that of control group (83.75% vs 61.25%,P < 0.05 );Clinical adverse cardic events rates at 4 weeks of treatment group were obviously lower than those of control group (P < 0.01 ).No obvious adverse reactions related to the treatment group were observed.Condusion The serum hs-CRP level is in close relation with acute myocardial infarction,which is the risk factor.It is effective and safe of brevisapine injection in the treatment of acute myocardial infarction.The effects of brevisapine on acute myocardial infarction might be involved in decreasing the level of hs-CRP and inhibiting the vascular inflammatory reaction.

ABSTRACT

Objective The diolistic assay has been modified to make it simpler and more efficient in labeling neurons and glia. Methods CNS neurons and glial cells were labeled with DiI diolistic assay in fixed tissue and living brain slices of C57/B6J mice. Results The method allowed the visualization of the fine structure of neurons and glia including synaptic structures such as dendritic spines. Conclusion With the method, the labeling efficacy of cell's fine structure is improved, making it preferable for the analysis of dendritic spine. In addition, the ability to label the living neuron and glia will extend its application vastly.
